Plants in natural environments are often exposed to fluctuations in light intensity, and leaf‐level acclimation to light may be affected by those fluctuations. Concurrently, leaves acclimated to a given light climate can become progressively shaded as new leaves emerge and grow above them. Acclimation to shade alters characteristics such as photosynthetic capacity. To investigate the interaction of fluctuating light and progressive shading, we exposed three‐week old tomato (Solanum lycopersicum ) plants to either lightflecks or constant light intensities. Lightflecks of 20 s length and 1000 μmol m?2 s?1 peak intensity were applied every 5 min for 16 h per day, for 3 weeks. Lightfleck and constant light treatments received identical daily light sums (15.2 mol m?2 day?1). Photosynthesis was monitored in leaves 2 and 4 (counting from the bottom) during canopy development throughout the experiment. Several dynamic and steady‐state characteristics of photosynthesis became enhanced by fluctuating light when leaves were partially shaded by the upper canopy, but much less so when they were fully exposed to lightflecks. This was the case for CO2‐saturated photosynthesis rates in leaves 2 and 4 growing under lightflecks 14 days into the treatment period. Also, leaf 2 of plants in the lightfleck treatment showed significantly faster rates of photosynthetic induction when exposed to a stepwise change in light intensity on day 15. As the plants grew larger and these leaves became increasingly shaded, acclimation of leaf‐level photosynthesis to lightflecks disappeared. These results highlight continuous acclimation of leaf photosynthesis to changing light conditions inside developing canopies.  相似文献

Background

Tsetse flies (Diptera: Glossinidae) are the vectors of African trypanosomosis, the causal agent of sleeping sickness in humans and nagana in animals. Glossina fuscipes fuscipes is one of the most important tsetse vectors of sleeping sickness, particularly in Central Africa. Due to the development of resistance of the trypanosomes to the commonly used trypanocidal drugs and the lack of effective vaccines, vector control approaches remain the most effective strategies for sustainable management of those diseases. The Sterile Insect Technique (SIT) is an effective, environment-friendly method for the management of tsetse flies in the context of area-wide integrated pest management programs (AW-IPM). This technique relies on the mass-production of the target insect, its sterilization with ionizing radiation and the release of sterile males in the target area where they will mate with wild females and induce sterility in the native population. It has been shown that Glossina pallidipes salivary gland hypertrophy virus (GpSGHV) infection causes a decrease in fecundity and fertility hampering the maintenance of colonies of the tsetse fly G. pallidipes. This virus has also been detected in different species of tsetse files. In this study, we evaluated the impact of GpSGHV on the performance of a colony of the heterologous host G. f. fuscipes, including the flies’ productivity, mortality, survival, flight propensity and mating ability and insemination rates.

Results

Even though GpSGHV infection did not induce SGH symptoms, it significantly reduced all examined parameters, except adult flight propensity and insemination rate.

Conclusion

These results emphasize the important role of GpSGHV management strategy in the maintenance of G. f. fuscipes colonies and the urgent need to implement measures to avoid virus infection, to ensure the optimal mass production of this tsetse species for use in AW-IPM programs with an SIT component.

Benzylisoquinoline alkaloids (BIAs) are a group of specialized metabolites found predominantly in the plant order Ranunculales. Approximately 2500 naturally occurring BIAs have been identified, many of which possess a variety of potent biological and pharmacological properties. The initial BIA skeleton is formed via condensation by a unique enzyme, norcoclaurine synthase, of the l-tyrosine derivatives dopamine and 4-hydroxyphenylacetaldehyde, yielding (S)-norcoclaurine as a central intermediate. The vast diversity of BIA structures is subsequently derived from (1) transformation of the basic BIA backbone by oxidative enzymes, particularly cytochromes P450 and FAD-linked oxidases, and (2) further structural and functional group modification by tailoring enzymes, which also include various reductases, dioxygenases, acetyltransferases, and carboxylesterases. Most of the biosynthetic enzymes responsible for the biosynthesis of major BIAs (i.e. morphine, noscapine, papaverine, and sanguinarine) in opium poppy (Papaver somniferum), and other compounds (e.g. berberine) in related plants, have been isolated and partially characterized. Diversity in BIA metabolism is driven by the modular and repetitive recruitment, and subsequent neo-functionalization, of a limited number of ancestral enzymes. In this review, BIA biosynthetic enzymes are discussed in the context of their respective families, facilitating exploration of common phylogeny and biochemical mechanisms.  相似文献

The Maugean skate (Zearaja maugeana) has only been recorded in two remote and isolated estuaries on the west coast of Tasmania, Australia. While the population status in one of these estuaries (Bathurst Harbour) is uncertain, it is likely that Macquarie Harbour now represents the sole remaining habitat for this species. Environmental conditions, in particular dissolved oxygen levels and benthic biodiversity, in Macquarie Harbour have deteriorated in recent years, impacted by increased nutrient inputs from an expanding salmonid aquaculture industry. These environmental changes are believed to pose a threat to the persistence of the Maugean skate. In assessing the risks for this rare and range-restricted species, it is vital to consider genetic information when developing management strategies. Both mitochondrial and microsatellite markers showed that the species has low genetic diversity; with no detectable genetic diversity in over 3000 base pairs surveyed from the mitochondrial genome, low average microsatellite heterozygosity (0.35?±?0.11), a low average number of alleles per locus (2.1?±?0.4) across eight microsatellite loci and no overall population structure within the microsatellite loci (Fst = ? 0.002, p?=?0.718?±?0.012). There was also evidence of a recent bottleneck or founder event, which may explain the low observed genetic diversity. While the species may have existed with low genetic diversity for many generations, the results of this study represent a flag for conservation concern for the Maugean skate. Given that Macquarie Harbour may be its last remaining habitat, any threats to the species resulting in local extinction could equate to global loss of this unique skate species.  相似文献

Declines in population size can compromise the viability of populations by reducing the effective population size (Ne), which may result in loss of genetic diversity and inbreeding. Temporal population genetic data can be a powerful tool for testing the presence and severity of reductions in Ne. The Gouldian finch (Erythrura gouldiae) is a flagship for conservation of Australian monsoonal savanna species. This species underwent severe population declines in the twentieth century due to land use changes associated with European colonization. Microsatellite and mitochondrial genetic data from Gouldian finch samples sourced from natural history collections prior to land use changes were compared with contemporary samples to estimate the severity of decline in effective population size and to detect changes in gene flow. These data show that Gouldian finch decline was not as severe as some sources suggest, and that population genetic connectivity has not changed following land use changes in the twentieth century. Multiple estimators of current Ne using genetic data from consecutive years suggest the Gouldian finch Ne is likely between a few hundred and a few thousand individuals, with some estimates within the range considered of conservation concern. This work has identified the need to genetically characterize populations in Queensland, and to understand critical demographic parameters (e.g. lifespan) in the Gouldian finch. Understanding these factors is vital to further improve genetic estimates of population size, key to the formation of appropriate conservation management of this species.  相似文献

Introduction

Efforts to harmonize lipidomic methodologies have been limited within the community. Here, we aimed to capitalize on the recent National Institute of Standards and Technology lipidomics interlaboratory comparison exercise by implementing a questionnaire that assessed current methodologies, quantitation strategies, standard operating procedures (SOPs), and quality control activities employed by the lipidomics community.

Objectives

Lipidomics is a rapidly developing field with diverse applications. At present, there are no community-vetted methods to assess measurement comparability or data quality. Thus, a major impetus of this questionnaire was to profile current efforts, highlight areas of need, and establish future objectives in an effort to harmonize lipidomics workflows.

Methods

The 54-question survey inquired about laboratory demographics, lipidomic methodologies and SOPs, analytical platforms, quantitation, reference materials, quality control procedures, and opinions regarding challenges existing within the community.

Results

A total of 125 laboratories participated in the questionnaire. A broad overview of results highlighted a wide methodological diversity within current lipidomic workflows. The impact of this diversity on lipid measurement and quantitation is currently unknown and needs to be explored further. While some laboratories do incorporate SOPs and quality control activities, these concepts have not been fully embraced by the community. The top five perceived challenges within the lipidomics community were a lack of standardization amongst methods/protocols, lack of lipid standards, software/data handling and quantification, and over-reporting/false positives.

Conclusion

The questionnaire provided an overview of current lipidomics methodologies and further promoted the need for community-accepted guidelines and protocols. The questionnaire also served as a platform to help determine and prioritize metrological issues to be investigated.

Biotic interactions have been controversial in distributional ecology, mainly in regards to whether they have effects over broad extents, with the negative view known as the Eltonian noise hypothesis (ENH). In this study, we evaluated the ENH for Phytotoma raimondii, a restricted‐range Peruvian endemic bird species: we developed models based on 1) only abiotic conditions, 2) only host plant distributions, and 3) both abiotic conditions and host plant distributions; models were evaluated with partial receiver operating characteristic test and Akaike information criteria metrics. We rejected the ENH for this case: biotic interactions improved the model. The frequency with which exceptions to the ENH are detected has important implications for distributional ecology and methods for estimating distributions of species.  相似文献

The Sirex woodwasp, Sirex noctilio (Hymenoptera: Siricidae) is considered a secondary pest of pine in its native range but has caused considerable economic losses in pine plantation forests in the southern hemisphere. In Brazil, trap trees are the primary tool used for early detection purposes but these are costly, labor-intensive to install and require stressing trees by herbicide application. Flight intercept traps baited with synthetic blends of host volatiles are an attractive alternative but have performed poorly in some settings. This study was carried out to look for alternatives to trap trees for use in Brazilian pine plantations for early detection of S. noctilio. Four field experiments were conducted in two consecutive flight seasons (2015–16 and 2016–17), in planted loblolly pine (Pinus taeda) stands, to compare captures among flight intercept traps baited with different lures, deployed at different heights and among different intercept trap designs. Two experiments compared different host volatile lures and a significant treatment effect was observed in one. No effect of trap design or height was observed.  相似文献
